Light-Induced Acid Generation on a Gatekeeper for Smart Nitric Oxide Delivery

Nitric Oxide (NO) is a next generation drug due to its participate in angiogenesis, apoptosis, immune responses, neurotransmission, and cardiovascular homeostasis in vivo. We report herein the design of a light-responsive gatekeeper for smart nitric oxide (NO) delivery by utilizing the calcium phosphate (CaP) coating layer and pH-jump reagent. This is the first system which imparts the pH- and light-responsiveness to the diazeniumdiolate which is extremely unstable under physiological conditions. In addition, its therapeutic potential on the wounded cornea has been demonstrated. This gatekeeper system provided knowledge for advancing the development of a superior delivery system of genes and drugs combined with NO.

우리 몸에서 혈관 팽창과 신경 전달, 면역반응, 상처치유 등에 작용하며 차세대 약물로 주목받고 있는 일산화질소를 빛에 의해서 전달할 수 있는 새로운 시스템을 보고하였다. 인화칼슘과 빛에 의해서 산을 내놓는 pH-jump reagen를 이용해서 개발된 문지기 (Gatekeeper) 시스템은 생체 내에서 불안정한 일산화질소 방출체에 산성 및 빛감응성을 부여하였으며, 개발된 시스템은 상처 난 각막을 효율적으로 치유할 수 있음을 확인하였다. 추후 혈관재협착, 연고제, 항암제 등의 다양한 분야로의 적용이 기대된다. 해당연구는 ACS Nano에 개제되었다.
